Cellobiose metabolism in Aerobacter aerogenes. 3. Cleavage of cellobiose monophosphate by a phospho- -glucosidase.

چکیده
An enzyme in extracts of Aerobacter aerogenes was shown to catalyze the hydrolytic cleavage of cellobiose monophosphate (6-0-phosphoryl-b-D-ghtcopyranosyl-(1 --f 4)-D-gillcase) to yield equimolar amounts of D-glucose and D-glucose 6-phosphate. The enzyme was partially purified and characterized. In addition to cellobiose monophosphate, it catalyzed the cleavage of cellobiitol phosphate, arbutin phosphate, salicin phosphate, gentiobiose phosphate, methyl P-D-glucoside phosphate, and phenyl P-D-glucoside phosphate, but not nonphosphorylated ,&glucosides.

متن کاملMetabolism of gentiobiose in Aerobacter aerogenes.
Cleavage of gentiobiose in cell extracts of gentiobiose-grown Aerobacter aerogenes was dependent on the presence of adenosine 5'-triphosphate (ATP). The enzymes that participate in the overall reaction were shown to be a beta-glucoside kinase, which catalyzes the phosphorylation of gentiobiose with ATP to form gentiobiose monophosphate [6-O-phosphoryl-beta-d-glucopyranosyl-(1 --> 6)-d-glucose],...
متن کاملCellobiose metabolism in Aerobacter aerogenes. II. Phosphorylation of cellobiose with adenosine 5'-triphosphate by a -glucoside kinase.
The first reaction in the metabolism of cellobiose in Aerobad er aerogenes was shown to be adenosine 5’-triphosphatedependent phosphorylation at C-6 of the nonreducing ring to yield 6-0-phosphoryl-O-D-glucopyranosyl-(1 + 4)-D-glucase (cellobiose monophosphate). The inducible fl-glucoside kinase that catalyzes the reaction was partially purified and characterized. In addition to cellobiose, the ...
متن کاملMutations that alter the regulation of the chb operon of Escherichia coli allow utilization of cellobiose.
Wild-type strains of Escherichia coli are normally unable to metabolize cellobiose. However, cellobiose-positive (Cel(+)) mutants arise upon prolonged incubation on media containing cellobiose as the sole carbon source. We show that the Cel(+) derivatives carry two classes of mutations that act concertedly to alter the regulation of the chb operon involved in the utilization of N,N'-diacetylchi...
متن کاملOvercoming inefficient cellobiose fermentation by cellobiose phosphorylase in the presence of xylose
BACKGROUND Cellobiose and xylose co-fermentation holds promise for efficiently producing biofuels from plant biomass. Cellobiose phosphorylase (CBP), an intracellular enzyme generally found in anaerobic bacteria, cleaves cellobiose to glucose and glucose-1-phosphate, providing energetic advantages under the anaerobic conditions required for large-scale biofuel production. However, the efficienc...
